Abstract

Background: Ibuprofen is anti-inflammatory nonsteroidal anti-inflammatory drugs 
(NSAID) from group derivative propionate acid which has activity antipyretic and 
analgesic. The Ibuprofen orally has side effect such as gastritis, dyspepsia, ulcers 
stomach or bleeding stomach. For to overcome this issue, it can used topical cream 
Because allow absorption drug directly to the areas in need, and reducing side effect 
systemic due to No pass channel digestion significantly. Objective: This study aims to 
optimizing the cream formula ibuprofen with variation emulator Tween 80 and Span 
80 as well known characteristics physicochemical optimum formula. Methods: Method 
optimization used is simplex lattice design by design expert. Optimization done with a 
variation comparison emulator Tween 80 and Span 80 for find out the optimal formula. 
Optimum formula preparation evaluated includes spreadibility, adhesion, pH, 
viscosity, and uniformity all. Data were analyzed using independent sample t-test 
(p<0.05). Results: Optimum formula own concentration Tween 80 is 8.5% and Span 80 
is 1.5%. Spreadibility test show results of 7±0.00 cm, while prediction from design 
expert of 6.89 cm. Result evaluation viscosity of 2800 ± 150.99 cps, while results 
prediction from design expert of 2716.31 cps so of that no significantly different. 
Conclusion:  Based on results it, concluded that method simplex lattice design proven 
effective for determine the optimum formula. Method This can be used optimally For 
determine the prediction formula the highest viscosity appropriate.
